Recherche de nouveaux gènes cibles dans le génome humain pour les récepteurs activés par les proliférateurs de péroxysomes (PPAR) : découverte du gène codant la sémaphorine 6B

Abstract : Peroxisome proliferator activated receptors (PPAR) are transcription factors. Their belong to the nuclear receptor superfamily and play roles in cell cycle as well as fatty acids and sugar metabolisms. Various PPAR agonists are known : unsaturated fatty acids, ecosanoïds and pharmaceutical compounds. Once PPAR is activated, it binds to retinoïc acid receptor (RXR). Thus the dimer is recruit on the PPAR's responsive element (PPRE). Sequences of known PPRE are made of two AGGTCA motif spaced by one nucleotide. We performed an immunoselection of PPRE sequence from human genomic DNA, in order to harvest new PPRE sequences (ISF). Sequence analysis of these ISF allow us to discover that a protein implicated in "axon guidance" called "semaphorin 6B" is a novel target gene for PPAR.
