Développement de systèmes de réservoirs de biomolécules à base de multicouches de polyélectrolytes

Abstract : We developed nanostructured thin polymeric films composed of multi strata to embed various active compounds. Polyelectrolytes are assembled by means of the "layer-by-layer" (LbL) method leading to the formation of polyelectrolyte multilayer films. Each reservoir is separated from the other by a barrier which prevent diffusion of molecules from one reservoir to the other one. Reservoirs are composed of biodegradable polyelectrolytes and barriers of non degradable polyelectrolytes or degradable polyesters. Inside the architectures, it is thus possible to embed one kind of biomolecules per reservoir. Bone rnarrow cells seeded on these films allow to test the sequential internalisation of embedded molecules : cells internalize first molecules located in the upper reservoir then in a second step they degrade the barrier and intemalize the molecules located in the lower reservoir. All kinetic parameters can be tuned by adjusting the total number of reservoir, the number of layers by reservoir, the thickness of the barrier or the degradability properties of the polymers that compose the banier. Confocal microscopy, atomic force microscopy, Raman spectroscopy, quartz crystal microbalance and fluorimetry are the main techniques used to characterize the different architectures and their efficiency. Two examples describe potential applications of these films i) anticancerous drugs can be embedded inside the films and cell accessibility to this dmg can be modulate, ii) nerve regeneration is possible using such films functionalized with growth factors which will be gradually release.
