Caractérisation du complexe multiprotéique comprenant le co-récepteur à l’acide Jasmonique JAZ6 de peuplier, cible de l’effecteur symbiotique MiSSP7 de Laccaria bicolor

Abstract : Establishment of ectomycorrhizal symbiosis between Populus trichocarpa and Laccaria bicolor requires the secretion of fungal small-secreted proteins named symbiotic effectors. One of these proteins MiSSP7 localizes in the nuclei of plant cells, where it binds PtJAZ6, a master regulator of the jasmonic acid signaling pathway. JAZ proteins of Arabidopsis thaliana form multiprotein complexes, containing in particular transcription factors. Formation of these complexes leads to the repression or induction of the genes regulated by the transcription factor. To understand the role of PtJAZ6, we aim to decipher the protein complex it belongs to. Using yeast two hybrid and BiFC experiments, we showed that PtJAZ6 interacts with the transcription factor PtJAM1 and JAM3 through its JAS domain. Furthermore PtJAM transcription factors are able to form homodimer and heterodimer with other PtJAM protein, but not with PtMYC2. TIFY domain of PtJAZ6 is needed for PtJAZ6 interaction with PtNINJA1 and PtNINJA3. These two proteins PtNINJA also interact with the three TOPLESS proteins, TRP4.1, TPR4.2, et TPL/TPR1.1. The TIFY domain is also required for the interaction of PtjAZ6 with MiSSP7. To infer the biological meaning of MiSSP7 binding to PtJAZ6 and its consequent effect on the transcription of PtJAM1-dependent genes, a bioinformatic study, still under ongoing improvement, was implemented.
