Purification du bactériophage MS2 et discrimination des phages infectieux et non infectieux par leurs propriétés de surface

Abstract : In case of viral contamination of environment, the determination of particles infectivity is essential for the health risk control. The aim of the present work was to provide information about viral particles surface properties (charge and hydrophobic/hydrophilic balance) to discriminate infectious and non-infectious virus. First of all, virus needed to be purified before further studies. Initially, purification of viral particles (MS2 phage) was evaluated by ion exchange chromatography on DEAE column. It led to a level of purity that seems close to dialysis but required more optimization. Secondly, to compare MS2 phages infectious and noninfectious, we used different methods: the ion exchange chromatography, the aggregation function of pH and adhesion to hydrophobic support. Of these three methods, chromatography provides information on these two types of particles, but still requires significant methodological development phases.
