Caractérisation de marqueurs d'identité cellulaire de la racine de riz

Abstract : I joined the DAR team (Développement Adaptatif du Riz, CIRAD, Montpellier) to do my internship. This group works on rice root development and specifically on genes involved in ground tissue formation. Some mutants under study in the lab are affected in cell identity (supernumerary cell layers or mixed cell identity). In order to be able to distinguish roots tissues one from the others and to analyze carefully these defects, the team needed cells identity markers. This tool was not available in the laboratory. Therefore, my project focused on characterizing such markers. I tested three different approaches, all based on cell wall composition. The first two ones rely on spectral signatures oftransverse section of rice seminal roots, which are excited at various wavelengths, such as UV (cell autofluorescence) and IR (FT-IR). The third approach was to screen a library of 174 antibodies specific to cell wall components by immunohistolocalization. At this stage of the project, we can conclude that the more promising results have been obtained by immunohistolocalization. Indeed, we succeeded by this method in identifying root tissue specific markers.
