Infections par des micro-organismes intracellulaires : approche biomoléculaire du diagnostic et de l'épidémiologie

Abstract : At the present time molecular biology is a fundamental tool in the research in biology. We have applied biomolecular techniques to the diagnosis and epidemiology of intracellular microorganisms. The first part of the manuscript present: 1) a comparative analysis of intracellular parasites and endosymbionts, and their involvement in the seriaI endosymbiotic theory on the origin of the eukaryotic cell ; 2) a review of the biomolecular techniques and their applications in microbiology. The second part is devoted to Chlamydia pneumoniae infections. We have developed a multiplex PCR assay for a rapid and differential diagnosis of C. pneumoniae and Mycoplasma pneumoniae respira tory infections. 2) Along with this study, an atypical chlamydial 165 rDNA has been detected, and phylogenetic analyses have indicated that is related to the parachlamydiae, recently associated to respira tory infections. 3) Il has been suggested that C. pneumoniae play a role in the atherosclerosis. We have searched for by RT-FCR chlamydial RNAs in atheromatous tissue speci'mens positive for the chlamydial DNA, in order to evidence viable organisms.This search has resulted negative. The third part focuses on molecular epidemiology of viral infections. 1) We have used the whole genome enzymatic restriction-based genotyping to show the stability in the time for Adenovirus 8 strains from ocular infections, and to reject the possibility of a common source of Adenovirus infections in a bone marrow transplantation unit. 2) We have analysed by nucleotide sequencing the variability of the X gene and the distribution of the genotypes of Hepatitis B Virus strains from chronic hepatitis or hepatocellular carcinoma patients.
