Etude de l'expression de la sex hormone-binding globulin (SHBG) dans les cellules de la granulosa humaines

Abstract : In this study, the expression of sex hormone-binding globulin (SHBG) in human granulosa cells has been investigated. First, the presence of SHBG has been detected in granulosa cells in situ, using immunohistochemistry (IHC) on adult and fetal ovarian sections. Second, the presence of the protein and the corresponding transcripts has been shown in primary granulosa cell cultures by IHC and RT-PCR. Furthermore, Western blot analysis of the culture medium demonstrated the secretion of SHBG by these cells and allowed to identify sorne regulatory factors, such as triiodothyronine and insulin. Finally, SHBG, in combination with estradiol, has been shown to act on the intracellular concentration of cyclic adenosine monophosphate and the progesterone secretion in cultured granulosa cells. These results suggest that locally produced SHBG may play a role in human follicular physiology.
