Déterminants nutritionnels et métaboliques de l'homocystéine au niveau hépatique : évaluation de 3 modèles expérimentaux : le rat gastrectomisé, thyroïdectomisé et/ou carencé

Abstract : Hyperhomocysteinemia can be associated with gastrectomy or thyroidectomy. However, pathologicals pathways and cellular effects remains unclear. We investigated 3 experimental models : gastrectomized rats, thyroidectomized rats subjected or not to methyl deficient diet, and pups from dam fed with deficient diet and submitted or not to transiet hypoxia. Gastrectomized rats exhibited hyperhomocysteinemia related to vitamin BI2 deficiency and low MTR activity. In this model, DNA methylation was correlated with MTR acivity and vitamine BI2 level, and was not with plasma homocysteine level. Thyroidectomized rats exhibited a drop of MTHFR and MTR activities related to decreased synthesis of vitamin B2 coenzymes. Unlike the human hypothyroid patient, these rats exhibited decreased total plasma homocysteine related to an increased activity of the hepatic trans-sulfuration enzymes (CBS). Pups born from dam fed with methyl deficient diet exhibited a dropfof MTR activity related to a real hepatic deficiency in vitamin BI2 related to an increased need during intrauterine life which was not compensated by the deficient diet during pregnancy.
