Adhésion leucocyte/endothélium : approches de l'interaction entre molécules d'adhésion LFA-1 et ICAM-1 par micropipette et chambre d'écoulement

Abstract : The aim of this work is to investigate the interaction of cell adhesion molecules LFA-1 and ICAM-1 by micropipette and flow chamber techniques. Two micropipettes hold respectively one SKW-3 (human T cell leukemia cell line) expressing LFA-1 and one red blood cell coated with soluble ICAM-1. Based on a stochastic model and a mechanical model of cell adhesion, dynamic parameters of adhesion are obtained. The firm adhesion of leukocytes on HUVEC under different shear stresses is studied by flow chamber. Experimental results show that the adhesion probality (Pa), the forward rate constant (kf) and the affinity (Ka), are increased with SKW-3 cells stimulated by Phytohemagglutinin (PHA) or ebdothelial cells stimulated by the cytokine TNF-[alpha], but tetramethylpyrazine (TMP) can significantly inhibit such increases. The Pa is significant higher for the lymphocytes of the patients with cerebral embolism. It is found that the Pa is similar between micro- and macro-vascular endothelial cells derived from same species (human), but that the Pa was different between microvascular endothelial cells isolated from different species (human and mouse).
