Conception et évaluation de biomatériaux polysaccharidiques pour le comblement de lésions ostéochondrales : Intérêt de l'association alginate-hyaluronate

Abstract : During traumatic chocs, cartilage defect become visible and its spontaneous repair evolved a fibrous cartilage. In this work, we have search to built a scaffold biomaterial to favoured the cartilage repair. For this, we have associated two polysaccharides: alginate for its structural properties and hyaluronate for its viscoelastic and biology properties. During the fir,st study, we have observed small interactions between the two polysaccharides in dilute and in concentrated solutions. The mixture of them in different proportions have permit to fotm hybrides matrices after ionic gelation. Rheologique measurement of the solide gel have schown a diminution of the hardness of the alginate gel when hyaluronate are introduced. The dea~etylation of hyaluronate, to reinforced the initial association, conducted in a depolymerization of the polysaccharide wich limited the interactions between the two polymers. The cytdcompatibility study, make with rats chondrocytes, have shown a benefique effect of hyaluronate on the proliferation and the viability of the cclIs. /\11 of the results permit us to purpose the use of middle viscosity alginate and high molecular weight hyaluronate, in proportion 1: 3, for the study of biocompatibility in rat experimental model of air pouch. This have confirmed a lack of inflanunation reaction coming from the hybrides matrices. It is then, possible to tb,ink that the gel can be used for an implantation in a model of animal cartilage defect.
