Cinétique, modélisation et amélioration de la production de L-lysine par Corynebacterium glutamicum sur milieu synthétique

Abstract : The microbial growth kinetics in batch and fed-batch culture, the improvement and the modelling of the lysine fermentation by C.glutamicum ATCC 21543 are presented. In the fist part, the influence of differents susbstrates (O2 , glucose and auxotrophic aminoacids) on growth, glucose consumption and lysine production is studied. The objectif of this kinetics analysis was first, to define process conditions for an improved lysine production, and second, to study the interaction between the consumptions of the substrates, growth and lysine synthesis. The results show that high glucose, O2 and auxotrophic aminoacids severely reduce the rate of growth uptake and production. On the other side, the deficiencies of auxotrophic aminoacids increase lysine yield (Yp/s) but reduc specific growth rate, glucose consumption and lysine productivity. Then, we defined a mode of operation of fed-batch process for an improved lysine productivity. To control the feeding we utilize a “software sensor” based on an indirect measurements (% CO2 an estimation algorithm and a process model ; Good performances are achieved. Finally, in the third part, based on the results onf the kinetics studies, a model is established for the fermentation process. The model is able to correctly simulate growth rate, substrates uptake (glucose, auxotrophic aminoacids, O2 and products formation (lysine and CO2) in batch and fed-batch culture over a wide range of experimental conditions
