Culture d'hybridomes en réacteur agité : eétudes cinétiques, modélisation et estimateurs mathématiques

Abstract : With the general objective of animal cell culture processes improvement, a study of hybridoma cultures in stirred bioreactor is realized following three steps : 1)Kinetic studies allowing the determination of the main metabolic schemes including the osmolality influence and the use of LDH as a cellular death physiological marker are made to give quantitative data on the studied strain. 2) A kinetic model describing medium composition and cell density evolutions usable in batch and continuous cultures is determined. 3) Medium composition and cell density evaluations from the measurement of one or two variables are realized coupling the model to mathematical estimation techniques like Extended Kalman Filter. The application of such methods is a prerequisite for further development of animal cell culture command strategies and reactor optimization
