Étude des excrétions carbonées et azotées de la microalgue tisochrysis lutea en conditions limitantes de nitrate ou de phosphate

Abstract : Variations of phytoplankton environment cause modifications on cellular metabolism but also on population evolution. To better understand the ecosystem dynamics, it is fundamental to study acclimatization capacity in response to environment and mineral composition variation, in particular regarding major nutrients such as carbon, nitrogen and phosphorus. Therefore this study aimed at evaluating dynamics for dissolved organic nitrogen (DON) and carbon (DOC) in N or P-limited batch culture of T-iso. Amino acids were identified and quantified by HPLC and D-glucose was quantified by colorimetric reaction with a 0.6 µM sensitivity threshold. P and especially N limitation cause glucose secretion. Under limited N, amino acid concentration is low and constant between 150 nM, while under limited P, microalgae secreted amino acids with concentrations that reached 2 µM. Nitrogen from amino acid nitrogen in the NOD fraction is twice as high under limited P as under limited N. Thus, this study validates the hypothesis that P limitation causes DON secretion. Nevertheless it was found that amino acids and glucose contribute little to DOC and/or DON fractions and do not explain major variations for the two fractions.
