Caractérisation des facteurs de transcription responsables de la virulence chez Toxoplasma gondii

Abstract : Toxoplasma gondii encodes several factors of virulence and invasion which are critical for its survival. The expression of these factors is closely monitored during the cell cycle of the parasite to allow their correct packaging into secretory organelles. However, little information is available on the factors that control the expression of these genes. In this context, we decided to characterize the AP2X-5 protein which belongs to the to the AP2 family of transcription factors. Our results of Immuno-bloting have confirmed that AP2X-5 interacts with another factor AP2, AP2XI-5, which was shown to be involved in regulating expression of virulence factors. Chromatin imùmunoprecipitation analysis allowed us to observe an enrichment of the AP2X-5 proteinat the level of promoters of genes coding for rhoptry proteins, as well as the promoter of the AP2XI-5 gene. Using immunofluorescence, the AP2X-5 protein expression was observed between the S and M phases of the cell cycle but not the G1 phase. These data indicates that AP2X-5 may play a role in regulating expression of important virulence factors.
