Évaluation par marqueurs moléculaires microsatellites de la diversité d’une collection d’aulnes glutineux provenant de Wallonie (Belgique), de Lorraine (France) et du Grand-Duché du Luxembourg

Abstract : Fourteen microsatellite markers were used to identify genotypes in a collection of 149 black alders (Alnus glutinosa) from an area covering Wallonia in Belgium, Lorraine in France and the Grand Duchy of Luxembourg. The objectives were to provide a tool for managing the collection and tracing the trees, to characterize black alder diversity in the region and to compare the diversity of the collection with the diversity of a population from the same region. The probability of identity varied from 7.5 10-1to 6.2 10-3, depending on the marker used, with a combined probability of 1.410-14for the 14 markers used simultaneously. These markers therefore proved to be a powerful tool for distinguishing black alder genotypes. Three of the markers were enough to discriminate 99 coppiced alder stools from a riverbank and highlight visual identification errors within these coppices. The microsatellite markers revealed high allelic diversity, with 176 alleles detected in the collection and the number of alleles per microsatellite locus ranging from 2 to 31. The observed heterozygosity of the collection ranged from 0.149 to 0.845, depending on the marker. An analysis of an alder population (25 trees tested) from the same region using the same microsatellite markers revealed close similarity among the most frequent alleles and no significant difference in frequency between the clonal collection and the population. This result showed that the allelic diversity of the collection was representative of a natural population from the same region.
