Evolution des compétences morphogénétiques des cultures de pin Maritime (Pinus pinaster Ait.) obtenues par embryogenèse somatique, et ses conséquences sur l'efficacité de ce système de multiplication végétative

Abstract : Due to rare transitions from immature to cotyledonary embryos, P. pinaster somatic embryogenesis yet remains uncompetitive as a mass propagation system. We first investigated factors affecting morphogenetic capacities of immature embryos during proliferation step. Second, evolution of maturation process has been qualitatively and quantitatively studied for one performant line, PN519. ABA and other plant growth regulators actions on maturation were finally analyzed in order to improve knowledge regarding this developmental transition. Morphogenetic ability of immature embryos which varied according to lines, decreased as a function of subcultures, in disconnection with their proliferation ability. Cell line origin definitely remained the main endogenous factor affecting morphogenetic ability. Nevertheless, weekly subculturing on a hormon-free and maltose-containing medium, reduced such degeneration. In line PN519, immature embryos developped differentially according to the embryonal-suspensor mass (ESM) they belonged to. Maturations occurred since immature embryos displayed organized cellular aggregation. Differentiations appeared promoted when using fluridone (ABA biosynthesis inhibitor) into the maturation medium. An endogenous ABA synthesis is suspected into ESM, that could interact with the exogenous application. Cotyledonary embryos length shortened according to the decrease of immature embryo quality. The disappearance of suspensor formation was suspected to be involved in such phenomenon. We concluded that proliferating somatic embryos should ideally be kept in a juvenile, synchronous and maturation-inducible stage, to enable mass production of P. pinaster emblings.
