Relation entre la distribution intracellulaire et les photodommages induits par la méta-tétra (hydroxyphényl) chlorine : effet du traitement photodynamique sur des cellules d'adénocarcinome mammaire humain de phénotype MDR

Abstract : Photodynamic therapy (FDT) is a rapidly growing modality for the treatment of light-accessible tumors. The PDT efficacy is influenced by the photochemical activity and the intracellular localization of the photosensitizers used. The study of meta-tetra(hydroxyphenyl)chlorin intracellular distribution revealed that the endoplasmic reticulum and the Golgi apparatus are the preferential sites of this sensitizer localization in human mammary adenocarcinoma MCF-7 and MCF-7/DXR cells. The photoinduced diminution of the activities of enzymes located in these two organelles observed immediately after irradiation revealed that the endoplasmic reticulum and the Golgi apparatus are the intracellular sites inilially damaged by the treatment. Whereas only few mTHPC localizes in mitochondria and mitochondrial enzymes are not affected by the treatmenr, the irradiation of cells sensitized with mTHFC induced a depolarization of the mitochondrial membrane and a release of cytochrome c from mitcchondria to the cytosol, These mitochondrial events seem to be implicated in MCF-7 cell death 24h after irradiation. The effect of mTHPC-mediated PDT on human adenocarcinoma cell lines has revealed that the MCF-7IDXR cells resistant to doxorubicin mediated chemotherapy and expressing the MDR phenotype were sensitive to PDT. MTHPC-mediated PDT could also be considered as an efficient way for destruction of neoplastic MDR cells.
