Corrélation croisée entre mécanisme dipolaire et anisotropie de déplacement chimique : dynamique des protéines en solution

Abstract : This manuscript deals with the determination and the exploitation of nitrogen-15 relaxation rates in labeled proteins. In the first part, we introduce the phenomenological aspect the heteronuclear relaxation. The second chapter is devoted to the measurement of these relaxation parameters in macromolecules. We present new methods to measure the cross correlation term coming from the interference between the dipolar (NH) and the nitrogen-15 chemical shift anisotropy relaxation mechanisms. The third chapter turns on the manner of extracting structural and dynamic information from these parameters. The strategy is based on a residue per residue analysis of the multifield nitrogen-15 relaxation; this approach is shown to be compatible with the assessment of anisotropic overall reorientation in proteins. The interest to include the cross correlation parameters in order to determine simultaneously the chemical exchange and the chemical shift anisotropy is also exposed.
