Influence de la contrainte de cisaillement sur la phosphorylation et la distribution de la protéine VASP dans les cellules endothéliales

Abstract : Vasodilator-stimulated phosphoprotein (VASP) is a member of the proline rich Ena/VASP protein family that links the cell membrane proteins, signal transduction pathways, and the actin cytoskeleton. VASP is associated, in different cell types, with micro filaments, focal adhesions, cell-cell contacts, and highly dynamic membrane regions. It is supposed to have an important role in the regulation of actin dynamics. ln this study the expression, redistribution, and the phosphorylation of VASP under flow conditions were examined. After treatment by shear stresses, a double labeling was used to visualize the relationship between the actin organisation and the distribution of VASP. Western blotting was used to detect the phosphorylation of VASP. Under different shear stresses, VASP in different cell types (ECV304, primary HUVEC, HUVEC cellline) always located at the ends of stress fibers. The shear stress of O.2Pa elevated the quantity of the phosphorylated VASP, reached a peak and then decreased. The shear stress of 1.OPa bas similar tendency as that of O.2Pa. On the contrary, the shear stress of 1.5Pa caused a lower level phosphorylation of VASP. These results show that shear stress induces the phosphorylation of VASP in endotheliale cells, and this phosphorylation depends on the intensity and the duration of shear stress.
