Recherche et étude de nouveaux chélateurs de l'aluminium et du phosphore utilisables en médecine

Abstract : In the first part of this study, the potential for the chelation of aluminum by different compounds has been first determined using two in vitro technics. The formation of stable complexes with aluminum in aqueous solution has been evaluated by using pulse polarography. This technique allows the influence of temperature and of calcium to be studied for each compound. An ultrafiltration technique combined with aluminum determination by atomic emission spectroscopy has allowed the ability of different substances to complex aluminum that was previously bound to serum proteins, to be estimated. The kinetics of chelation and the minimum efficient concentration have been determined for all the products studied. The real efficacies of the compound were studied by in vivo investigations to compare the effectiveness of the best chelating agents on the distribution and excretion of aluminum, after repeated intraperitoneal administrations to rats. The aim of the second part was to highlight possible new nonaluminum phosphate-binders to limit hyperphosphatemia in patient with renal failure. So, different compounds was evaluated as dietary phosphate binders in rats. Aluminum chloride was evaluated as a reference. Animals were divided in different groups, receiving several doses of the different substances. All along the treatment, urines and stool were collected. At the end of the treatment, animals were sacrified and plasma and different organs were collected. Concentrations of phosphorus and calcium in all samples were determined. The possible transfer of the substances in rat tissue was verified
