Potentiel de transformation de souches d'intérêt de Streptococcus salivarius pour l'étude du transfert d'Eléments Intégratifs et Conjugatifs

Abstract : Streptococcus salivarius is a human commensal bacteria that can be an opportunistic pathogen. Genomic sequencing of selected strains allowed us to highlight the presence of mobile genetic elements type Integrative and Conjugative Elements (ICE). They carry accessory loci that could confer selective advantage to its host (antibiotics or phage resistance or pathogenicity properties). In order to study ICE transfer between different strains of S. salivarius, a reporter system based on a set of plasmid construction was developed. During my M2 project I studied, in silico and in vitro, the potential of natural transformation (competence) of S. salivarius strains of interest to labeled the ICE for transfer experiments. As streptococci competence depend on quorum-sensing system, different times and quantities of signal peptide and DNA were tested. To increase competence level, recovery kinetic was done to fix the time for foreign DNA integration and expression. All of these experiments allowed us to measure competence level of S. salivarius interest strains.
