Étude comparative des méthodes de détection des adénovirus infectieux aux stades précoces de l’infection

Abstract : Adenoviruses belong to the group of the pathogenic enteric viruses and are most commonly found in aquatic environments. Adenoviruses are responsible for wide range of diseases, including respiratory infections, conjunctivitis and gastroenteritis. In this study we aimed to asses and to compare different cell culture based methods of detection of infectious adenoviruses. Two different in situ methods have been analysed: the detection of the viral protein hexon by immunostaining and the detection of the hexon mRNA using the molecular beacon. The third method is the ICC-PCR based on the real time PCR quantification of the viral genome in the infected cell. The comparison of the methods is based on the sensitivity and the rapidity of the detection. The PCR and the immunofluorescence methods applied to the epifluorescence microscopy led to the detection of the adenoviral infection as early as 14h post-infection.
