Développement et validation de modèles adaptés à la mesure de l'anxiété et du stress chez le rat Wistar : application à la caractérisation des effets protecteurs d'un hydrolysat trypsique de caséines bovines enrichies en caséine [alpha]s1

Abstract : Currently, the pharmacological care of anxiety- and stress-related disorders presents a limited efficacy, and then justifies the need for developing new treatments. In this context, the properties of a tryptic hydrolysate from bovine caseins (CTH) are of interest and remain to be clarified. For that, this work aimed in a first part to define the most sensitive conditions for detecting the effects of potent anxiolytic compounds in the well-known behavioural model of anxiety, the elevated plus-maze (EPM), and to develop both acute and chronic stress models able to modify anxiety- and depression-like behaviour in rat. In a second part, these models have been applied to the evaluation of the anxiolytic and antistress putative properties of CTH when orally administrated. The results indicated that 1) the anxiety level of rats tested in the plus-maze is related to methodological parameters (luminosity, closed wall type, extramaze space), and 2) the CTH shows in behavioural tests as the EPM and the conditioned defensive burying an anxiolytic activity that is different from diazepam used as a reference. The results also indicated that 1) the physiological and behavioural responses to an acute stress exposure depend on the model of stress (restraint or forced swim), and 2) the CTH reduces both stress induce responses. Finally, two models of chronic stress have been developed. The CTH has been tested only in one model that consists in submitting rats to chronic variable stresses.In these conditions, the CTH reduced the behavioural hyperactivity induced by the chronic stress exposure, but not the stress-related physiological alterations. In conclusion, this study confirms the anxiolytic and antistress properties of the CTH when orally administrated and suggests a mechanism of action that may be different from that of benzodiazepines.
