Etude du métabolisme carboné chez Streptomyces pristinaespiralis :

Abstract : During this study, pristinamycins production showed variability and reproduction troubles. When a culture was made following the previous experimental procedures but using a different bioreactor, no pristinamycins production was observed. After experimentations on agitation speed, medium composition and inoculum age effect, a new procedure for batch reactor cultures was set. Surprisingly, in these conditions, pristinamycins production occurred during growth phase and without apparent nutritional limitation. Glucose consumption is high, and organic acids as succinic acid and lactic acid were poorly excreted. During these cultures, the redox potential decreased while the pristinamycins production occurred. This could be useful for cultures monitoring. Enzymatic activities of the central metabolism and intracellular pH (pHi) were also assayed during these cultures. Taking into account the pHi for the specific activities led to few differences from the assayed values. The pentoses-phosphate pathway seemed to be important for pristinamycins synthesis. This is in accordance with its reduced co-factors regeneration activity, such as NADPH, which are necessary for the pristinamycins production pathway
