Caractérisation fonctionnelle du complexe ribonucléoprotéique TRBP/RPAP3

Résumé

The TAR RNA Binding Protein (TRBP) is a key element in several pathways, including regulation of HIV-1 replication and infection, or miRNA maturation in the cytoplasm by the RNase III Dicer. For the first time, we identified a connection between the RNA Polymerase 2 Associated Protein 3 (RPAP3) of the R2TP complex and TRBP, which directly interact together. Our preliminary results showed that the interaction between TRBP and RPAP3 was mutually exclusive from that between TRBP and Dicer. We have consequently observed that RPAP3 could modulate miRNA function, probably through a TRBP sequestration mechanism and competition for Dicer binding. Preliminary results are also indicative of a possible role in regulation of HIV translation, through impaired binding of TRBP to the TAR element on the genomic RNA. Our project will therefore answer important questions regarding the role of RPAP3 in the regulation of TRBP activities. This is all the more so exciting and original as this connection between TRBP and RPAP3/R2TP has never been described, neither regarding the function of TRBP during HIV replication, nor regarding its role in miRNA processing.
